Abstract:This paper is to establish the optimum extraction technology of total polyphenols(TP) in Clematis manshurica Rupr. by ultrasonic extraction method. By using the extraction efficiency of TP in Clematis manshurica Rupr. as evaluating indicator, some important factors, such as the concentration of ethanol, the extracting time, the ratio between raw material and solvent and the extracting times, are optimized by orthogonal experimental design. The content of TP in Clematis manshurica Rupr. is determined by spectrum photometric method at detection wavelength of 766nm. The optimum extraction condition of TP in Clematis manshurica Rupr. with ultrasonic method are obtained as follows: the concentration of ethanol, the extracting time is 35min, the ratio of raw material to solvent is 1∶30, and the extraction times is 3times is 55%. The concentration of gallic acid and its absorption value shows a good linear relationship at the range of 1.116~11.160μg/mL. It is concluded that the ultrasonic extraction method obtained in the paper is easy to operate and reliable, so it could be used for extracting TP in Clematis manshurica Rupr.
